Brief Det~nitive Report Dw/ld-related Molecular Polymorphism of Dr4 Fl-chains*

چکیده
Antigenic determinants encoded by genes of the HLA-D region have been related to at least three separate molecular complexes, DR, MB (also known as DC and LBE), and SB. At the cell surface each complex has a dimeric structure composed of a structurally conserved a-chain noncovalently associated with a polymorphic fl-chain. The DR and MB series of determinants have been extensively characterized using allospecific serological reagents and, to date, 10 DR and 3 MB allotypes are recognized. Beyond these serologically detectable polymorphisms, HLA-D region products exhibit an additional degree of antigenic complexity revealed by cellular reagents such as homozygous typing cells (HTC). These cells respond to antigenic determinants (Dw/LD determinants) that are phenotypically distinct from those recognized by available antisera. Thus, within a panel of individuals of a single serologically defined DR type, such as DR4, a number of different D w /LD specificities including Dw4, Dwl0, DYT, DB3, and LD40 exist (1). In this report we have addressed this cellularly detected antigenic complexity from a molecular perspective. Using isoelectric focusing (IEF) to analyze DR molecules immunoprecipitated from a panel of individuals homozygous for DR4, but expressing different Dw/LD determinants, we have observed that (a) between DR4 fl-chains, but not a-chains, considerable structural polymorphism exists and (b) the particular DR fl-chain structural configuration observed upon IEF is associated with the cell Dw/LD type. These data, thus, provide the first molecular evidence associating the Dw/LD determinants recognized by cellular reagents to the serologically defined DR molecule.

Dw/LD-related molecular polymorphism of DR4 β-chains
The results of this study demonstrate the existence of molecular heterogeneity (polymorphism) within DR beta-chains isolated from a single serologically defined DR phenotype, DR4. The data are consistent with the possibility that this polymorphism is related to the Dw/LD phenotype as defined with the cellular reagents, homozygous typing cells.
متن کاملTwo-dimensional gel electrophoretic analysis of the MT3 and DR4 molecules from the different D-typed cells
This report demonstrates directly, using two-dimensional gel electrophoresis and alloantisera, the following: (a) The DR4 light chains show a structural polymorphism among the Dw4, DKT2, and DYT cells. (b) Most of the class II light chains consist of the DR light chain. (c) The MT3 molecule is distinct from the DR4 molecule in the Dw4, DKT2, and DYT cells. (d) The MT3 molecule does not show any...
متن کاملBrief Det~nitive Report Intragenic Recombination in an Ea Gene for a Murine Ia Antigen*
T he plasma membranelocated murine Ia antigens consist of two polymorphic chains: the larger chain, designated a, is ~35,000 mol wt, whereas the smaller chain, designated fi, is ~28,000 mol wt (1). Whereas the I-A antigens are encoded by genes (Aa and AO) located within the I-A subregion, the I-E antigens are encoded by one gene (E~) located in the I-E subregion and a second (Ep or A,) 1 in the...

Recently (1), we described a new genetic system that is HLA-DQ-related . Two alleles, 2B3 and TA10, can be recognized serologically . The 2133 specificity detected with the 11133 mAb was previously described (2) as a DQwl-related specificity . The TA 10 specificity is DQw3-related and was first. described by Maeda (3) with the TA 10 mAb. TA10 can also be detected with alloantisera (4) . Recentl...
متن کاملPrimary association of HLA-DQw8 with type I diabetes in DR4 patients.
DNA from 164 Caucasian type I (insulin-dependent) diabetic patients and 200 Caucasian nondiabetic control blood donors were analyzed by the polymerase chain reaction technique for HLA-DR4 and the associated Dw and DQB subtypes of DR4. The DQw8 subtype of HLA-DR4 was associated with type I diabetes in all DR4 subgroups (DR4/3 and DR4/non-3). Dw subtypes of DR4 other than DW10 did not confer addi...
